Dates (Phoenix dactylifera L.) from the Adrar region of Algeria are rich in polyphenols, and ternary solvent extracts’ antioxidant activity correlates with condensed tannins content

Abstract


Dates are the leading fruit consumed by native population of the Algerian Sahara. This study aimed to assess the phytochemical contents and antioxidant activities of twelve date palm cultivars (Phoenix dactylifera L.) flesh grown in the Algerian Sahara. Crude extracts were obtained by macerating samples in methanol/acetone/water (MAW, 7/7/6, V/V/V) at room temperature. Phenolics, flavonoids, and condensed tannins were determined in crude extracts, and the antioxidant capacity of last was evaluated by DPPH, ABTS•+, and FRAP assays. Our results shown that total phenolics ranged between 97.75 and 421.15 mg GAE/100 g DW, total flavonoids between 14.46 and 59.53 mg CE/100 g DW, and condensed tannins (polymeric proanthocyanidins) between 21.90 and 76.42 mg CE/100 g DW. Fractionation of the aqueous phase from binary solvent mixtures (80% methanol) showed that n-butanol phase recorded highest flavonoids content (up to 35.34 mg CE/100 g DW) while the residual aqueous phase of 80% acetone recorded highest content of condensed tannins (up to 78.38 mg CE/100 g DW). DPPH assay revealed IC50 values ranged between 0.026 and 0.181 mg/mL of tested extracts. Moreover, the antioxidant activity of these extracts ranged between 865.19 and 1815.16 µmol TE/100 g DW and between 863.50 and 1980.46 µmol TE/100 g DW when assessed by ABTS and FRAP assays, respectively. High antioxidant capacity of MAW extracts is correlated mainly with condensed tannins content. We conclude that studied dates are good sources of phenolic compounds, and ternary solvent mixture extracts exert noticeable antioxidant activities.
