Gladiolus hybrida L. is an economically important species belonging to Iridaceae family. Owing to its huge market demand globally, mass multiplication of novel cultivars of this species is desirable. Application of micropropagation techniques for ornamental plants are being applied in the agricultural industry, ensuring true-to-the-type clones of superior genotypes. Despite its tremendous potential in floriculture, very few studies have been carried out to optimize the tissue culture protocol supporting commercial farming of Gladiolus in India. meta-Topolin (mT) is an adenine-based cytokinin which enhances shoot proliferation and plant development during in-vitro culture. The aim of the current study was to assess the effect of different concentrations of mT on in-vitro propagation of two commercially important Gladiolus varieties, namely Darshan and Arka Pratham. In this study, four different concentrations of mT supplemented media were compared to standard MS-media for evaluating the in-vitro growth parameters. Corms harvested from field-grown plants were used as explants. MS media supplemented with 2 mg/l mT exhibited significantly higher and healthier shoot proliferation than all other treatments. Two different combinations of NAA and IAA were further assessed for in-vitro rhizogenesis, where shoots induced from 2 mg/l mT media yielded the highest results in MS media + 2.0 mg/l NAA + 1 mg/l BAP. Arka Pratham consistently performed better than Darshan. Survivability of > 80% was observed in the plantlets after 15 days of primary hardening. The findings of our study establish an optimized mT-based in-vitro propagation technique, establishing it as a superior alternative for enhancing commercial Gladiolus production.
