The B3 gene family is a group of transcription factors that play important roles in the regulation of plant growth and development, as well as in the response to biotic and abiotic stresses. This gene family has been well studied in various plant species but in C. melo, it is unknown. In the current study, a total of 54 B3 genes were identified with the comprehensive genome-wide identification studies and were classified into four classes namely, ARF, REM, LAV and RAV. The physicochemical feature analysis revealed that the protein is hydrophilic in nature and centrally localized in the nucleus. The gene architecture and motif analysis revealed the class specific pattern. The phylogenetic analysis showed class specific clades based on their sequence similarity. REM and ARF class were two major clades. The gene expression profiling showed the highest expression of CmARF2 and CmARF9 genes under fruit developmental stage whereas under biotic stress CmARF2, CmARF9 and CmARF14, CmLAV3, CmLAV6, CmRAV2 and CmRAV7 genes were highly upregulated. The current study provides a platform for the functional characterization of potential melon B3 genes under fruit developmental stage as well as against diverse biotic and abiotic stress at the molecular level.
