The effect of explant type and plant growth regulators on determining the pathway of organogenesis in Brachystelma glabrum was evaluated. Leaf, internode, node and TCL of node explants were evaluated for their ability to induce either direct or indirect organogenesis. Murashige and Skoog (MS) medium supplemented with different cytokinins, auxins and organic supplements either alone or in combination for their effect on direct or indirect organogenesis. In direct organogenesis, TDZ (2.0 mg/l) produced maximum number of direct shoots regeneration from leaf and TCL of node explants and they were further multiplied on TDZ (2.0 mg/l) with NAA (0.5 mg/l). Indirect organogenesis was achieved through organogenic callus from leaf, internode and node explants. Best response of callus induction was achieved on medium fortified with NAA (0.5 mg/l), and further shoot regeneration from callus was recorded on TDZ (0.5 mg/l). Multiplication of shoots was maximum on TDZ (0.5 mg/l) and NAA (0.5 mg/l) combination. Coconut water (CW) supplementation at 25% further enhanced the shoot quantity in both direct and indirect organogenesis. The microshoots of both direct and indirect organogenesis were rooted on half-strength MS medium incorporated with NAA (0.5 mg/l) and the hardening showed 70% of survival. Among the explants used, leaf explant found to be best for both direct and indirect pathway of organogenesis. This is the first report on direct and indirect organogenesis of Brachystelma glabrum. These two regeneration systems will be useful for the germplasm conservation of Brachystelma and future genetic transformation and functional genomics research.
