In this study an attempt has made to develop reliable and reproducible in vitro regeneration protocol to get healthy and well-formed plants from shoot tip, cotyledonary node hypocotyls and leaves of pomegranate cv. Bhagava for transformation and tissue culture. Shoot tips cultured on full MS+BAP (2.0 mg l−1) showed best response (70%) to shooting. Cotyledonary nodes cultured on full MS + NAA (2.0 mg l−1) + BAP (2.0 mg l−1) showed 80% shooting with 4–5 shoots per explant. Highest (60%) embryogenic callus from hypocotyl explants was induced on full MS + 2, 4-D (0.5 mg l−1) + kinetin (0.5 mg l−1). For rooting plantlets were cultured on half MS + activated charcoal (500 mg l−1). Well rooted plantlets of pomegranate cv. Bhagava acclimatized on the medium containing sand: soil: FYM (1:1:1) and kept in the greenhouse.
