The present investigation is an effort to establish callus formation, preliminary phytochemical studies and antibacterial activity of methanolic extracts of callus of Holarrhena antidysenterica. Which is an important medicinal plant of great economic value in the Indian subcontitent and continue to provide valuable therapeutic uses. Explants used for callus formation were mainly seeds and in vivo and in vitro grown plant parts like stem, nodal parts, roots and apical shoots. This study involves the preliminary phytochemical screening, and separation of compounds present in the crude extracts of callus by TLC and HPTLC method. Dry powder of callus was used for qualitative analysis of phytochemicals and found to contain phenolic compounds, alkaloids, amino acids, tannins, anthraquinone glycosides, flavonoid. Presence of peaks in the TLC and HPTLC exhibited presence of principle bioactive compound conessine in the methanolic extracts of callus. Methanolic extracts of callus was tested for antimicrobial activity against gram positive and gram negative bacteria. The microorganisms tested were staphylococcus aureus, Salmonella typhi and Escherchia coli.
