Although Dalbergia sissoo Roxb. is an important tree species that help in nitrogen fixation, it lacks distinguishable morphological markers. There are only a few shisham breeding studies available. In this study, the molecular diversity of 30 shisham genotypes collected from different regions of India and one foreign landrace obtained from Nepal were analyzed using a randomly amplified polymorphic DNA (RAPD) marker. The genetic diversity of shisham genotypes were assessed by calculating various marker parameters, namely percent and number of monomorphic and polymorphic loci. This gave an average of 8.76 bands per primer, of which 65 bands (24.71%) were monomorphic for all the genotypes, and 198 bands (75.28%) were found to be polymorphic for one or more genotypes. Jaccard’s coefficient of similarity between all possible pairs of genotypes revealed that genotypes had divergence ranging between 22.4 and 49.8%. The phylogenetic results showed that group I, II, III, and IV comprise genotypes originating from plains of U.P., while genotypes in groups III and IV were from hills and foothills of Uttarakhand state. However, PS-71, PS-8 and Ps-26 were exceptional. The RAPD marker system aided in determining genetic diversity, origin, and identifying variation among landraces.
