The essential oil was extracted from the aerial parts of Santolina africana Jord. & Fourr. (Asteraceae) using hydrodistillation and then analyzed by gas chromatography–mass spectrometry (GC–MS). In total, 96 compounds accounting for 100% of the total oil. The essential oil (EO) is distinguished by its high content of oxygenated monoterpenes (38.19%) and hydrocarbon monoterpenes (34.33%). The chemical composition of the essential oil derived from the aerial parts was determined to be as follows: bicycle [3.1.1] heptane,6,6-dimethyl-2-methylene (18.39%), eucalyptol (15.78%), 2(10)-pinene (10.67%), and (-)-germacrene D (5.88%). Furthermore, the methanol extract (ME) of the aerial parts showed notable antioxidant activity according to the DPPH free radical scavenging with an IC50 value of 72.33 ± 0.15 µg/mL. The highest levels of antibacterial activity were demonstrated by the MeOH extracts of the aerial parts, with inhibition zone diameters of 21 ± 1 mm against Escherichia coli ATCC 8739 at 16 mg/ml and of 13 ± 1 mm against Staphylococcus aureus ATCC 25,923 at 16 mg/ml.
