Potato (Solanum tuberosum L.) (Solanaceae family) is the third most important field crop, having more than 4000 varieties and cultivars including over 180 wild species. The cultivar Tezu local is a nutrient-rich indigenous potato but cultivated mostly in north-eastern India. To conserve and commercialize this cultivar, an inclusive microclonal propagation protocol was developed in this study. Tuber sprouts were initially cultured in Murashige and Skoog (MS) media supplemented with 0.5, 1.0, or 1.5 mg/L kinetin, meta-Topolin (mT), or zeatin. As many as 5.0 shoots were initiated from single sprout in 0.5 mg/L mT-supplemented MS medium. In vitro shoots were transferred for rooting in MS media supplemented with 1.0, 2.0, or 3.0 mg/L indole-3-acetic acid or indole-3-butyric acid (IBA). The maximum number of roots (20.7) were induced with 3.0 mg/L IBA. Mono-phasic (simultaneous) shoot and root development was achieved with a combination of mT (0.5, 1.0, or 1.5 mg/L) and 3.0 mg/L IBA, wherein the combination of 1.0 mg/L mT and 3.0 mg/L IBA produced the most promising results. The plants, thus produced, were hardened first in the cocopeat, where they showed 100% survival. Upon transfer from cocopeat to field conditions, 85% plant survival was recorded. Flow cytometry analysis confirmed the unaltered ploidy level of the plants. Genetic fidelity analysis via polymerase chain reaction with start codon-targeted polymorphism, inter simple sequence repeats, directed amplification of minisatellite-region DNA, and conserved DNA-derived polymorphism primers revealed monomorphic banding profiles. The field growth of micropropagated plants inside the shade net house was accomplished. This protocol can be evaluated to propagate (in large-scale), conserve, and commercialize other indigenous potato cultivars.
