Abstract


The present investigation was undertaken to characterize the mitochondrial DNA (mtDNA) of cytoplasmic male sterile line 185A, maintainer line 185B, restorer line RSLG 580R and hybrid 185A × RSLG 580R of sorghum using 40 RAPD and 16 ISSR primers. Out of which 23 RAPD and 12 ISSR primers showed the polymorphism in CMS system of sorghum. The random primer OPC 20 amplified the product of ∼2.84 kb size only in CMS line and hybrid of the sorghum which confirms that cytoplasmic male sterility is maternally inherited trait. Further, it was observed that 12 RAPD and 08 ISSR primers synthesized unique fragments only in mtDNA of CMS line i.e. 185A and were absent in its maintainer, restorer lines and hybrid which might be associated with CMS phenotype of 185A in sorghum. The results of present study may have great usage in sorghum breeding.
