Sanghuangporus has long been recognized as a valuable medicinal mushroom due to its various bioactive properties, including antitumour, antioxidant, anticancer, and antimicrobial activities. This study aims to collect and identify Sanghuangporus strains from Vietnam to serve as source materials for the development of Sanghuangporus-based medicinal products. During the study, a wild strain of Sanghuangporus, collected from Bach Thao Park, Hanoi, was confirmed as a new record of Sanghuangporus vaninii based on both morphological characteristics and phylogenetic analysis. To enhance the production of polysaccharides and β-glucans, we optimized the cultivation conditions for surface fermentation of this Sanghuangporus vaninii. The results indicated that PDA was the most suitable medium for mycelial growth. Yeast extract and soluble starch were identified as the optimal nitrogen and carbon sources, respectively. The optimal pH was determined to be 8.0, with an incubation temperature of 30°C and the mycelium harvesting time was five weeks. Under these optimized conditions, the dry mycelium weight reached 6.98 g/L, while polysaccharide and β-glucan yields were 5.20 g/L and 0.96 g/L, respectively.
