Cleome spinosa Jacq. is ornamental plant of family Cleomaceae having horizontal racemes of white or pink flowers, 5–7 digitate, numerous brown seeds in siliqua, attaining height of 1–1.5 m. Cleome gyandra Linn. another plant of this family is annual, herbaceous weed growing on waste land having 3–5 foliate leaves. Literature reveals both plants having active ingredients, possessing medicinal properties.
Tissue culture was performed to study callus induction and regeneration potential of both the plants. Stem segments and leaves, taken as explants were cultured on MS media containing various combinations of Auxins and Cytokinins viz. 2,4-D, NAA, IBA, IAA, Kinetin and BAP. Buds regenerated from stem segments on plain MS medium and also on MS+NAA (0.1mg/l-1.0mg/l) and BAP (0.5mg/l-2.5mg/l). In these combinations callus formation was noted on stem and leaf explants. Callus induction on leaves was 100% at lower concentrations of NAA (0.1mg/l-1.0mg/l) and kinetin or BAP at the concentrations of 0.5mg/l-2.5mg/l. When callus was sub-cultured on MS+ Kinetin (0.1mg/l-0.5mg/l) and BAP (0.5mg/l-1.5mg/l), 1–3 shoots regenerated per explant. MS+IBA (0.5mg/l-1.5mg/l) resulted in production of few roots on 50% of shoot buds. Callus induction and regeneration potential was found more in Cleome spinosa Jacq. than in Cleome gynandra Linn.
