Micropropagation response of three grape vines (Vitis vinifera L.) cultivars to ferrous, nitrate and growth regulators

Abstract


Three Grapevine (Vitis vinifera L.) cultivars, Kamaly, Taify and Rash Mew were propagated by tissue culture technique and a reproducible and reliable protocol was developed through this investigation. Single nodes of grapevine were initiated on MS medium after getting healthy aseptic cultures by their surface sterilization with 0.15% HgCl2 for 6 minutes. The shoots were multiplied by the addition of BA, Fe and N. The best multiplication parameters were obtained from the Kamaly cultivar grown on MS medium supplemented with 3.0 mgl-1 BA and 10 mgl−1 Fe. Whereas, the culture of Taify grape cultivar on MS medium supplemented with 10 mgl−1 N produced the longest shoots (3.00 cm). The best rooting results were obtained for Taify and Rash Mew cultivars especially when grown on full MS salts strength media supplemented with 1.0 mgl−1 NAA. A high survival rate of gradually acclimatized Rash Mew plantlets was obtained (100%). The best culture mixture for acclimatization stage was composed of peatmoss and perlite.
